It would be Weber's boldest move by far, significantly expanding the company's US presence while boosting its position in rare diseases, gastrointestinal disorders and neuroscience, where Shire is a leader in drugs to treat hyperactivity.
Buying Shire would be transformational for Takeda but would be a big financial stretch, since the company is worth in excess of $10 billion more than the Japanese group.
Takeda's reported offer comes amid a flurry of transactions in the pharmaceutical sector, marked by GlaxoSmithKline Plc's $13 billion agreement last month to buy out Novartis AG's stake in the two companies' consumer-health joint venture.

Shire also rejected the third proposal, made up of £17.75 in cash, to be paid in U.S. dollars, and £28.75 of new Takeda shares. That would value Shire at approximately 43 billion pounds, or $61 billion, Thomson Reuters data showed. The first valued Shire at up to £44.00 per share, the second was £45.60 and the third was £46.50.
The UK's Shire has said it has rejected a bid of around £42 billion, around $60 billion, from Takeda, but is in talks with the Japanese firm's advisers to secure a better deal.
In reaction, Shire's shares jumped 8% to £40.70 each in London afternoon trading.
A bidding war for Shire is threatening to break out after Botox maker Allergan said it was considering a takeover offer for the rare diseases drug maker.
This week Shire said it was selling its cancer treatment unit to France's Servier for £1.7bn in a deal that was initiated in December and was unrelated to the possibility of a takeover bid.
